A medical report issued by the club on Friday afternoon confirmed a thigh problem for Vazquez, who would have been in line for a start against Arandina in the Copa del Rey this weekend.
«Following tests carried out today by the Real Madrid Medical Services on our player Lucas Vazquez, he has been diagnosed with an injury to the semitendinosus muscle in his right thigh. His progress will be monitored,» read a statement on Real Madrid's official website.
The 32-year-old has made 20 appearances for Real Madrid this season, scoring once and providing three assists, and he is expected to be out for at least the next three weeks.
As a result, Vazquez will miss the game with Arandina, in addition to the Spanish Super Cup in Saudi Arabia, where Los Blancos will meet Atletico Madrid in the semi-finals ahead of a potential final against Barcelona or Osasuna.
A three-week absence would also see him miss out against Almeria in La Liga on January 21 before potentially returning against Las Palmas on January 28.
Kroos, meanwhile, suffered an ankle issue during Real Madrid's 1-0 win over Mallorca on Wednesday, and the German is not involved in the squad against Arandina.
However, head coach Carlo Ancelotti is hopeful that the experienced midfielder will be available for the Spanish Super Cup.
